Overview

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the ACTH(4-7) fragment, to which a Pro-Gly-Pro tripeptide confers enzymatic stability. It is devoid of corticotropic activity.

Mechanism of action

In research, Semax increases BDNF and NGF expression (CREB pathway), modulates dopaminergic and serotonergic neurotransmission, and displays neuroprotective properties. In the rat hippocampus, a single administration was associated with an approximately 1.4× increase in BDNF and 1.6× in TrkB phosphorylation.

Research context

Semax is approved in Russia for ischaemic stroke and certain cognitive disorders. It is approved neither in the EU nor in the United States.
